On our network, we have a standalone HP fax machine which is network capable and on a separate floor. It’s a pain having to save my faxes, go up two floors, fax and return to my desk.

What I am wanting to do is use Winfax from my Desktop and get it to fax using the networked fax machine two floors above.

Is this possible?

no, this isn’t possible but you can install a fax modem on your PC and connect it to a phone line for faxing, then you can fax directly from your PC using WinFax.
